Fat Digestion: Emulsification by Bile and Lipase

中2 · 生物単元: 消化と吸収(酵素のはたらき)Fat Digestion: Emulsification by Bile and Lipase 目安 15分about 15 min
Students understand that bile does not break down fat but only splits it into small droplets (emulsification), and that the larger surface area lets lipase efficiently break fat into fatty acids and glycerol, which are then absorbed by the villi of the small intestine.

Predict (before you touch anything)

What do you think happens to the large fat droplets in the small intestine when bile is added?💡 胆汁には消化酵素が含まれていないことを思い出しましょう。Remember that bile contains no digestive enzymes.
If there is no bile and only a lot of lipase is released, do you think the fat will be digested well?💡 リパーゼは脂肪の粒のどこにくっついてはたらくのかを考えましょう。Think about which part of a fat droplet lipase has to attach to in order to work.

Try it

Keep "Bile" at 0, set "Lipase" to 100 and watch for about 10 seconds. Note how the purple lipase bounces off the large fat droplets, and record the "Fat left" percentage and the "Large drops" value under "Lipase digestion speed".
Press "Reset", then set "Lipase" to 0 and "Bile" to 100. Watch the big droplets split into small ones when the green bile hits them, and check that "Fat left" stays at 100% even as the number of "small (emulsified)" droplets increases.
With Bile still at 100, raise "Lipase" to 100. Watch lipase attach to the small droplets, releasing orange fatty acids and light-blue glycerol that are absorbed into the villi of the small intestine at the bottom.
Wait about 30 seconds and record how the "Fat left", "Digested, moving" and "Absorbed by villi" percentages change, along with the "Small drops" digestion speed.

Check

With lipase but no bile, how much of the fat was digested?→ ほとんど分解されない。 Almost none. Even after about 10 seconds "Fat left" stays at 100%, and the digestion speed for large drops is only about 0-1%. Lipase bounces off the big droplets.
When only bile was added, what happened to the number of fat droplets and the "Fat left" percentage?→ 粒の数は 3 個から 30 個前後に増えたが、「脂肪のまま」は 100% のまま。 The number of droplets rose from 3 to around 30, but "Fat left" stayed at 100%. Bile only splits the droplets; the amount of fat does not go down (it is not digested).
With both Bile and Lipase at 100, what happened to the fat?→ 乳化した小さな粒が次々に分解され、乳化後(小)の分解の速さは 30〜50% 程度になった。 The small emulsified droplets were digested one after another, with a "Small drops" digestion speed of roughly 30-50%. After about 25 seconds "Absorbed by villi" had risen to about 40-60% and "Fat left" kept falling.
What substances were produced by digestion, and where did they go?→ 脂肪酸(オレンジ色)とグリセリン(水色)ができ、小腸の壁の絨毛へ吸収された。Fatty acids (orange) and glycerol (light blue) were produced and absorbed into the villi of the small intestine wall.

Think further

Bile contains no digestive enzymes, so why is it said to "help digest fat"?→ 胆汁が脂肪を細かい粒にすると、同じ量の脂肪でも表面積が大きくなる。 When bile breaks fat into tiny droplets, the same amount of fat has a much larger surface area. Lipase can only work on a droplet's surface, so more surface means more places for lipase to attach, and digestion goes faster.
Powdered sugar dissolves in water faster than a sugar cube. What does this have in common with lipase working faster after emulsification?→ どちらも物質を細かくして表面積を大きくし、まわりの水や酵素とふれる面を増やすことで、変化を速めている点。In both cases, breaking the substance into smaller pieces increases its surface area, so more of it touches the surrounding water or enzyme, which speeds up the change.

Bile is made in the liver and stored in the gallbladder; lipase is in pancreatic juice from the pancreas. The sharp switch in lipase efficiency by droplet size is a simplification for visibility; in reality digestion speed changes continuously with surface area. Also, the actual products are fatty acids and monoglycerides, which are rebuilt into fat inside the villi and enter the lymph vessels; this model only shows up to absorption.
