Starch ➡ Maltose (How Amylase Works)
Students understand that salivary amylase, a digestive enzyme, cuts starch (a chain of glucose units) into maltose (two units each) while remaining unchanged and working again and again, and that it works fastest around body temperature (about 37°C), hardly works at low temperature, and is permanently denatured at high temperature (about 60°C or above).

Predict (before you touch anything)
After amylase breaks down starch, do you think it gets used up, or does it remain afterwards?💡 だ液はほんの少しでも、たくさんのデンプンを分解できることを思い出しましょう。Remember that even a small amount of saliva can break down a lot of starch.
If test tubes of starch solution mixed with saliva are kept in ice water (about 5°C), at body temperature (37°C), and in hot water (about 70°C), how do you think the speed of breakdown will compare? And what if the hot tube is later cooled back to 37°C?💡 酵素はタンパク質でできています。 Enzymes are made of protein. Think about what happens to the protein in an egg when it is heated.
Try it
In the starting state ("Temperature" 37°C, "Amylase" 3), watch what happens when a green amylase bumps into a yellow starch chain: the end of the chain fits into the pocket, the bond breaks, and a pink maltose (2 units) moves away. Use "Pause" to freeze and "Start" to resume if needed.
Drag the "Time" slider to 5:00 to fast-forward, then read "Starch left", "Maltose" and "Amylase" in the display and look at the graph.
Press "Reset", set "Temperature" to 5°C and then 20°C, fast-forward "Time" to 5:00 each time, and compare the percentage of "Starch left" with the 37°C result.
Press "Reset", let some breakdown happen at "Temperature" 37°C, then raise "Temperature" to 70°C and watch the enzyme shapes and the display. Next set "Temperature" back to 37°C, wait a while, and check whether breakdown starts again.
Press "Reset" and, at "Temperature" 37°C, set "Amylase" to 1 and then 6, comparing "Starch left" at "Time" 1:00 in each case.
Check
How did the starch change as it was broken down? Did it fall apart into single glucose units?→ 鎖の端から2個ずつ切り取られ、鎖が短くなりながらピンクの麦芽糖(2個分)がふえていった。 Two units at a time were cut off the end of a chain, so the chains got shorter while the number of pink maltose molecules (2 units each) grew. It never broke into single glucose units (48 units of starch can give at most 24 maltose molecules).
After fast-forwarding to 5:00, had the number or shape of the amylase molecules changed?→ アミラーゼは3個のまま「正常」で、形も変わらず、何度もデンプンの鎖を切り続けていた。There were still 3 amylase molecules, all "normal" and unchanged in shape, and they kept cutting starch chains again and again.
At 5:00, roughly how much "Starch left" was there at 37°C, 20°C and 5°C?→ 37°Cでは残り約0〜20%まで減ってほとんど分解され、20°Cでは約40〜80%が残って37°Cより明らかに遅く、5°Cでは100%のままでほとんど進まなかった(分子の動きは乱数なので、毎回の数値はかなりばらつく)。At 37°C it fell to about 0–20%, so almost all was broken down; at 20°C about 40–80% remained, clearly slower than at 37°C; at 5°C it stayed at 100% and almost nothing happened (molecular motion is random, so the numbers vary quite a lot between runs).
After heating to 70°C and returning to 37°C, did the breakdown start again?→ 70°Cにするとアミラーゼのくぼみがつぶれて「すべて変性」と表示され、分解が止まった。 At 70°C the amylase pockets collapsed, the display showed "all denatured", and breakdown stopped. Back at 37°C the "Starch left" percentage did not change, so breakdown did not restart. Only after "Reset" (new enzymes) did it work again.
Think further
How is the fact that human digestive enzymes work best around 37°C related to the body? And what problem might a long-lasting very high fever cause?→ ヒトの体温はほぼ37°Cに保たれているので、その温度で酵素が最もよくはたらき、消化がすばやく進む。 Human body temperature is kept at about 37°C, so enzymes work best at that temperature and digestion goes quickly. If body temperature gets too high, enzymes and other proteins can be permanently denatured, which could damage body functions.
Why did breakdown get faster when the number of amylase molecules was increased from 3 to 6?→ アミラーゼがデンプンの鎖にぶつかって結びついたときに分解が起こるので、酵素の数が多いほど一定時間にぶつかる回数がふえ、同時に切れる鎖もふえるから。Breakdown happens when an amylase molecule bumps into and binds to a starch chain, so with more enzymes there are more collisions in the same time and more chains can be cut at once.
Common misconceptions
The idea that an enzyme is used up in the reaction. In fact amylase is the same in shape and number before and after, and works over and over again.
The idea that higher temperature always makes an enzyme work better, or that cooling restores an enzyme damaged by heat. Activity peaks near body temperature, and once denatured above about 60°C the enzyme does not recover when cooled.

Molecular motion is random, so the numbers vary slightly from run to run even with the same settings (have students compare several runs). The model is simplified: starch is shown as six unbranched 8-unit chains, cut two units at a time from the end. Real amylase also cuts within chains, and other enzymes in the small intestine break maltose down into glucose. One second on screen corresponds to five seconds of the experiment.
