Thylakoid Membrane: Light Reactions Simulator

Students understand that in the light reactions on the thylakoid membrane, PSII and PSI absorb light and pass electrons from water to NADP⁺, that H⁺ builds up in the lumen along the way, that ATP is made as this H⁺ flows back to the stroma through ATP synthase, and that the rate depends on light intensity and on how well chlorophyll absorbs the wavelength.

Predict (before you touch anything)

If blue (450 nm), green (550 nm) and red (680 nm) light of the same intensity are shone on the membrane, which color do you think gives the weakest electron flow?💡 葉が何色に見えるかを思い出しましょう。 Recall what color leaves look. Was the light of the color that reaches your eyes absorbed by the leaf?
If the light is switched off, do you think ATP production stops immediately or continues for a while?💡 ATP合成酵素を回しているのは光そのものではなく、何の流れでしょうか。What actually turns ATP synthase: the light itself, or the flow of something else?

Try it

In the starting state ("① Light intensity" 70%, "② Wavelength" 450 nm, "③ Speed" 1.0×), watch for about 15 seconds: electrons (yellow dots) move PSII→PQ→b6f→PC→PSI→Fd→FNR, O₂ bubbles and H⁺ are released into the lumen at the OEC, and ATP synthase spins. Record "Lumen pH" and the "ATP total", "NADPH total" and "O₂ total" counters.
Move "② Wavelength" to 550 nm and observe "Absorption η(λ)", the (theory) value of "Electron input e⁻/s", the absorption curve at the top left, and green photons passing straight through the membrane into the lumen.
Set "② Wavelength" to 680 nm, read the same values, and compare them with 450 nm and 550 nm.
Return the wavelength to 450 nm, set "① Light intensity" to 100%, wait about 10 seconds, and record how "Lumen pH", "Lumen H⁺" and the production rates (/s) change.
Set "① Light intensity" to 0% and watch for about 20 seconds. Check in what order the NADPH and O₂ rates, the ATP rate and "Lumen pH" change. Use "Pause" to freeze the screen if needed.

Check

What was "Absorption η(λ)" at about 450 nm, 550 nm and 680 nm? What happened to the 550 nm photons?→ 450 nm で約 90%、550 nm で約 5%、680 nm で約 84%。 About 90% at 450 nm, about 5% at 550 nm and about 84% at 680 nm. Most 550 nm green photons were not absorbed and passed straight through the membrane, and the theoretical electron input dropped from 5.0 to about 0.3 e⁻/s.
On which side of the thylakoid membrane were O₂ and H⁺ released? On which side were NADPH and ATP made?→ 水の分解(2H₂O→O₂+4H⁺+4e⁻)によるO₂とH⁺はPSIIのルーメン(内腔)側に出た。 O₂ and H⁺ from water splitting (2H₂O→O₂+4H⁺+4e⁻) were released on the lumen side of PSII. NADP⁺+2H⁺+2e⁻→NADPH (at FNR) and ADP+Pᵢ→ATP (at ATP synthase) took place on the stroma side.
How did the lumen pH change when "① Light intensity" was set to 100%?→ pH The pH fell from about 7.2 at the start to about 6.2 at 70% and about 6.1 at 100%, and the number of lumen H⁺ icons increased. The electron input rose (theory 7.2 e⁻/s), so H⁺ built up faster from water splitting and b6f.
After light intensity was set to 0%, what stopped right away and what continued for a while?→ O₂ と NADPH の生成はすぐに減って 0 になったが、ATP の生成はルーメンにたまったH⁺が流れ出る間しばらく続いた。 O₂ and NADPH production dropped to zero quickly, but ATP production continued for a while as the stored H⁺ flowed out of the lumen. After about 20 seconds the pH had recovered to about 6.8 and ATP synthesis had almost stopped.

Think further

Why was ATP still made for a while after the light was switched off? What does ATP synthesis directly need?→ ATP合成酵素を回すのは光ではなく、ルーメンとストロマの間のH⁺の濃度差(勾配)だから。 ATP synthase is driven not by light but by the H⁺ concentration difference (gradient) between the lumen and the stroma. While the gradient built by light remains, H⁺ flows back to the stroma and ATP is made; when the gradient becomes small, it stops (chemiosmosis).
How is the fact that leaves look green related to the results of this simulation?→ 葉緑素は青と赤の光をよく吸収し、緑の光(550 nm 付近)をほとんど吸収しない。 Chlorophyll absorbs blue and red light well and absorbs very little green light (around 550 nm). The unabsorbed green light is reflected or transmitted and reaches our eyes, so leaves look green. Light that is not absorbed cannot drive electron transport, so photosynthesis is slow in green light.

The model is simplified: b6f moves 1 H⁺ per e⁻ (more in reality because of the Q cycle), 1 ATP = 4 H⁺, and one H⁺ icon in the lumen stands for about 2 H⁺. The measured "Electron input" value lags behind changes, so have students compare the (theory) value. At 700 nm the absorption is shown as about 23%, but the explanation text wrongly shows the "near green" message, so compare wavelengths at 450/550/680 nm.
