Why Doesn't the A Band Shorten?

生物 · 生物単元: 筋収縮(アクチンとミオシン)Why Doesn't the A Band Shorten? 目安 20分about 20 min
Students understand that in muscle contraction neither the thick (myosin) nor the thin (actin) filaments change length but only slide past each other, so the A band stays the same width while only the I band and H zone narrow, and that tension depends on how the filaments overlap.

Predict (before you touch anything)

When a muscle contracts and the sarcomere gets shorter, which of the A band, I band and H zone do you think keeps the same width?💡 それぞれのバンドが「どのフィラメントがある範囲」で決まるかを考えましょう。 Think about which filaments define each band. Do the filaments themselves shrink?
Do you think the force (tension) a muscle can produce keeps increasing the more the sarcomere is stretched?💡 力を出すのはミオシン頭部がアクチンをつかむ部分です。 Force comes from myosin heads gripping actin. What happens to that region if the sarcomere is stretched too far?

Try it

In the starting state ("Sarcomere length (target)" 2.20 μm, "Animation speed" ×1.0), watch the repeated contraction and relaxation and check whether the "A band", "I band" and "H zone" readouts on the right change. If it is too fast, lower the speed slider to about ×0.5.
Press "Pause (hold at target)" and move the "Sarcomere length (target)" slider to 3.60 → 2.60 → 2.00 μm. At each length, record the widths of the A band, I band and H zone and the "Relative tension".
Lower it further to 1.80 μm and 1.60 μm, and check the "Thin overlap" value, the orange overlap region in the middle, and the message "Stop: Z lines touch the thick filaments" shown at 1.60 μm.
In the "Length (μm)–tension" graph at the bottom right, check where the ● marker sits at each length and read off the range where tension is greatest.
Press "Back to rest length 2.2μm", then "Play (contract / relax)" to restart the animation, and confirm that the thin filaments themselves do not change length as the Z lines move toward the M line.

Check

When the sarcomere length was changed from 3.60 μm to 1.60 μm, what happened to the width of the A band?→ 常に 1.60 μm(「不変」)のままだった。 It stayed at 1.60 μm ("constant") the whole time. The A band is the region containing the thick filaments, and their length (1.6 μm) does not change.
What were the widths of the I band and H zone at 3.60 μm, 2.60 μm and 2.00 μm?→ Iバンドは 2.00 → 1.00 → 0.40 μm、Hゾーンは 1.60 → 0.60 → 0.00 μm と狭まった。 The I band narrowed 2.00 → 1.00 → 0.40 μm and the H zone 1.60 → 0.60 → 0.00 μm. When the sarcomere shortens by 1.0 μm, the I band also narrows by 1.0 μm.
What happened when the length went below 2.00 μm? The tips of the left and right thin filaments crossed the M line and began to overlap: "Thin overlap" was 0.20 μm at 1.80 μm and 0.40 μm at 1.60 μm. At 1.60 μm the I band became 0.00 μm and the Z lines touched the ends of the thick filaments, so it could not shorten any further.
At which lengths was the "Relative tension" greatest, and what was it at 3.60 μm?→ 2.0〜2.2 μm It reached 100 % around 2.0–2.2 μm. It was about 71 % at 2.60 μm and 0 % at 3.60 μm, and on the short side it dropped to about 90 % at 1.80 μm and about 80 % at 1.60 μm.

Think further

Why is the tension 0 % at a sarcomere length of 3.60 μm?→ 細いフィラメント(Z線から 1.0 μm)の先端がM線から 0.8 μm の位置まで離れ、太いフィラメントの端(M線から 0.8 μm)と重ならなくなるから。 The tips of the thin filaments (1.0 μm from the Z line) end up 0.8 μm from the M line, which is exactly where the thick filaments end (0.8 μm from the M line), so they no longer overlap. Myosin heads cannot grip actin, no cross-bridges form, and no force is produced.
When myofibrils are observed under a microscope, what mechanism is supported by the fact that the A band width does not change during contraction?→ フィラメントが縮むのではなく、アクチンとミオシンが互いに滑り込むことで短縮するという「滑り説(滑走説)」の証拠になる。 It supports the sliding filament theory: the sarcomere shortens because actin and myosin slide past each other, not because the filaments shrink. If the filaments shrank, the A band would also get narrower.

The model uses simple geometry with 1.6 μm thick filaments and 1.0 μm thin filaments, and shows the H zone as the region with no thin filaments (= sarcomere length − 2.0 μm). So the thin filaments reach the M line at 2.0 μm, and at 1.6 μm the Z lines hit the thick filaments and shortening stops (also explained in the on-screen "Model assumptions"). The drop in tension on the short side of the curve is an approximation to experimental data, and the myosin head motion is schematic.
