Muscle Contraction: Sliding of Actin and Myosin

生物 · 生物単元: 筋収縮(アクチンとミオシン)Muscle Contraction: Sliding of Actin and Myosin 目安 20分about 20 min
Students understand that in muscle contraction myosin heads use an ATP-driven chemical cycle to pull actin filaments toward the M line, so the filaments slide past each other without changing length: the Z lines move closer and only the I band and H zone shorten, while the A band stays constant.

Predict (before you touch anything)

When a muscle contracts, which of the A band, I band, and H zone in the sarcomere do you think change length, and which stay the same?💡 A帯は何のフィラメントがある範囲か、I帯とH帯はそれぞれ何が「ない」範囲かを思い出しましょう。Recall which filament defines the A band, and what is missing from the I band and from the H zone.
If Ca²⁺ is present but ATP is completely gone, do you think the muscle will relax or become stiff?💡 ATPがミオシン頭部のサイクルのどの段階で必要になるかを考えましょう。Think about at which stage of the myosin head cycle ATP is needed.

Try it

With the default settings ("(1) Ca²⁺ level" 5.0 µM, "(2) ATP level" 3.0 mM, "(3) Speed" ×1.0), watch for about 30 seconds and record how the "Sarcomere length", "H zone edges" (top right) and the band bar (I band, A band, H zone) change. Also confirm that the ringed head in the close-up goes through steps ① to ⑤ on the right.
Press "Reset", let it contract for about 10 seconds, then set "(1) Ca²⁺ level" to 0 µM. Look at the tropomyosin (blue strand) and troponin (dots), the "heads on actin" count, the sarcomere length, and the status line under the close-up.
Return "(1) Ca²⁺ level" to 5.0 µM so contraction starts, then set "(2) ATP level" to 0 mM partway through. Observe the sarcomere length and the number of "rigor (A·M) heads".
Keeping ATP at 0 mM, lower "(1) Ca²⁺ level" to 0 µM and check whether the length recovers; then return "(2) ATP level" to 3.0 mM and see what happens.
If the motion is too fast, lower "(3) Speed" to about ×0.3 and confirm in the close-up that right after Pi is released, the head angle falls from 90° to 45°.

Check

As contraction proceeded with the default settings, how did the sarcomere length, H zone, I band, and A band change?→ 全長は 2.60 µm から少しずつ短くなり、約30秒で 2.00 µm に達した。 The length shortened gradually from 2.60 µm and reached 2.00 µm after about 30 s. The H zone width went from 0.60 µm to 0 ("gone"), and the I band (both sides combined) shrank from 1.00 µm to 0.40 µm. The A band stayed at 1.60 µm (fixed) the whole time.
When the Ca²⁺ level was set to 0 µM, what happened to the heads and the tropomyosin?→ トロポニンCa²⁺結合率が 0%、開放された結合部位が 0/48 になり、トロポミオシンが結合部位を覆った。 Troponin with Ca²⁺ dropped to 0% and open sites to 0/48, so tropomyosin covered the binding sites. Heads on actin fell to 0 and power strokes to 0/s. The close-up showed "Low Ca²⁺: tropomyosin blocks binding sites", and the sarcomere returned toward its resting length of 2.6 µm (relaxation).
What happened when ATP was set to 0 mM during contraction? Did the length recover when Ca²⁺ was then set to 0?→ 重なり部分の頭部がすべて(例: 32/32)アクチンに結合したまま「硬直(A·M)」状態になり、状態表示は「硬直(リガー):ATPなしで頭部が外れない」。 All heads in the overlap (e.g. 32/32) stayed bound to actin in the "rigor (A·M)" state, and the status read "Rigor: no ATP, heads cannot let go". The length froze at its current value (e.g. about 2.45 µm) and did not recover even when Ca²⁺ was set to 0. When ATP was returned to 3.0 mM, the heads detached and, with Ca²⁺ at 0, the sarcomere relaxed to resting length.
In the close-up, release of what triggers the power stroke, and what does ATP do?→ Pi Release of Pi makes the head tilt from 90° to 45° (power stroke), moving actin toward the M line. ADP is then released, giving the rigor bond; ATP binding detaches the head from actin, and hydrolysis of ATP (to ADP·Pi) re-cocks the head to 90°, ready to bind again.

Think further

Using what you saw in the simulation, explain why muscles become stiff some time after death (rigor mortis).→ 死後は ATP が作られなくなる。 After death, ATP is no longer made. Myosin heads need ATP to bind before they can detach from actin, so without ATP they stay locked onto actin in the rigor state; the filaments cannot slide and the muscle becomes stiff. In the simulation, with ATP = 0 all heads stayed bound and the length did not change.
When the sarcomere reached 2.00 µm the H zone disappeared and it stopped shortening. Each thin filament is 1.0 µm long, so at a total length of 2.0 µm the tips of the left and right actin filaments meet at the M line and the H zone (the region without actin) vanishes. Neither actin nor myosin changes length, so in this model no further sliding is possible; the heads keep cycling but the length stays the same (isometric tension).

The number of heads and the distance per stroke are simplified for visibility, and relaxation assumes titin springs the sarcomere back to its 2.6 µm resting length. Troponin's Ca²⁺ binding is computed with a Hill equation (K = 1 µM), giving about 50% at 1 µM. Point out that during rigor, lowering Ca²⁺ changes the tropomyosin display but the heads still do not detach, to summarize that relaxation needs both low Ca²⁺ and ATP.
