Stomata Up Close: How Guard Cells Open and Close
Students understand that a stoma opens when its guard cells take in water and swell and closes when they lose water and shrink, and that the amount of transpiration depends on how wide the stomata are open and on the humidity of the outside air.
Open the simulation
Predict (before you touch anything)
When the two guard cells around a stoma take in water and swell, do you think the stoma opens or closes?💡 保衛細胞は気孔に面した側のかべが厚くなっています。 The wall of a guard cell is thicker on the side facing the pore. When the cell swells, think about which side stretches more easily: the thin side or the thick side.
If the stomata are open by the same amount, on which kind of day do you think more water vapor leaves the leaf: a humid day or a dry day?💡 ぬれた洗たく物が早くかわくのはどんな日か思い出してみましょう。Think about what kind of day wet laundry dries fastest.
Try it
In the starting state (Light intensity 70%, Air humidity 40%, Water supply from roots 80%), look at the leaf cross-section and find the upper epidermis, palisade layer, spongy layer, vein (vascular bundle), lower epidermis, and where the stomata are. Also read the "Stoma opening", "Transpiration", and "Guard cells" displays below.
Slide "Light intensity" down to 0% and watch how the guard cells, the "Top view" inset at the lower right, and "Stoma opening" change. Then raise it to 100% and compare.
Keeping Light intensity at 100%, change "Air humidity" from 10% to 95% to 100%, and compare the number of light-blue dots (water vapor) leaving the stomata and the "Transpiration" display.
Set Air humidity back to 40%. With Light intensity still at 100%, set "Water supply from roots" to 0% and watch the stoma opening and the guard cells. Then set it to 40% and read how far the stomata open.
Check
Were the stomata in the upper epidermis or the lower epidermis of the leaf?→ 下表皮だけにありました(このシミュレーションでは3個)。 Only in the lower epidermis (three of them in this simulation). There are no stomata in the upper epidermis.
What happened to the guard cells and the stomata when Light intensity was 0%? At 0% the guard cells shrank, the stoma opening became 0%, and transpiration was "stopped". At 100% the guard cells swelled, the stoma opening reached 100%, and transpiration was "strong".
With Light intensity at 100%, what happened to the stoma opening and to transpiration as you raised Air humidity?→ 気孔開度は 100% のまま変わりませんでしたが、水蒸気の点は減り、蒸散の強さは 10% で「強い」、95% で「弱い」、100% で「停止」になりました。The stoma opening stayed at 100%, but there were fewer vapor dots: transpiration was "strong" at 10%, "weak" at 95%, and "stopped" at 100%.
When Water supply from roots was 0%, did the stomata open even with 100% light?→ 開きませんでした。 No. The guard cells stayed shrunken, the stoma opening was 0%, and transpiration was "stopped". At 40% supply the stomata opened only to about 50%.
Think further
When the soil is dry during a drought, what are the advantage and the disadvantage for a plant of closing its stomata?→ 利点は、蒸散で水が出ていくのをおさえて、しおれるのを防げることです。 The advantage is that it reduces water loss by transpiration and keeps the plant from wilting. The disadvantages are that the plant can take in less carbon dioxide through the stomata, so photosynthesis slows, and the pull that draws water up from the roots by transpiration becomes weaker.
Why do you think stomata are found mostly on the underside of the leaf (the lower epidermis)?→ 葉の裏側は直射日光が当たりにくく温度が上がりにくいので、必要以上に水が失われるのをおさえながら、気体の出入りができるからだと考えられます。The underside gets less direct sunlight and stays cooler, so the leaf can still exchange gases while avoiding losing more water than it needs to.
Common misconceptions
The idea that swelling guard cells block the pore. In fact, when they swell they bend outward and the gap between them (the stoma) opens.
The idea that an open stoma always releases the same amount of water vapor. When the outside air is humid, transpiration is low even if the stomata are open.

For teachers
Many Japanese textbooks call guard cells 孔辺細胞, so introduce both names. The model simplifies by setting the stoma opening from only light intensity × water supply, with humidity affecting only the transpiration rate (real stomata also respond to CO2 concentration, drought, and plant hormones, and some plants have stomata on the upper surface too).
