Water's Path: Tracing Water from Roots to Leaves with Red Dye

Water's Path: Tracing Water from Roots to Leaves with Red Dye 目安 20分about 20 min
Students understand that water absorbed by the roots rises only through the vessels (xylem) on the inner side of the stem's vascular bundles to the leaves and leaves through the stomata as water vapor, and that the more active transpiration is, the faster water is drawn up.

Predict (before you touch anything)

If you stand a white-flowered plant in water with red ink for a while and then slice its stem crosswise, which part do you think will be stained red?💡 茎の中には、水の通り道と、葉でつくられた養分の通り道があります。 Inside the stem there is a pathway for water and a separate pathway for food made in the leaves. Which one would the red water use?
How do you think the speed at which a plant draws up water differs between a windy day and a humid, still day?💡 葉から水が出ていく量と、根から吸い上げる量の関係を考えてみましょう。Think about how the amount of water leaving the leaves is related to the amount taken up by the roots.

Try it

In the starting state (Wind strength medium (50), Air humidity 50% (normal), Open stomata 70%), follow the red "Tracking" particle in "(b) Cross-section [zoom]" as it goes root hair → cortex → vessel (xylem). Also check where the Xylem (inside) and Phloem (outside) are in the "Stem cross-section".
Press "View: whole" (or tap the picture) to switch to the whole view. Watch red particles rise through the stem's vessels into the leaf vein (xylem) and leave from stomata on the underside of the leaf as blue particles (water vapor). Also confirm that the blue box in (a) Outside view is the part enlarged in (b).
Move "Wind strength" to 0 and 100, and "Air humidity" to 0% and 100% (keeping the others at their starting values), and record the "Transpiration" (particles/min) and "Water rising speed" (%) values.
Set "Open stomata" to 100% and to 0%, and observe Transpiration and the blue particles leaving the stomata.
Finally, compare Transpiration and Water rising speed for the combination Wind strength 100, Air humidity 0%, Open stomata 100% and the combination Wind strength 0, Air humidity 100%, Open stomata 0%.

Check

Where in the stem did the red particles (red water) travel? Did any rise through the phloem or the epidermis?→ 赤い粒は維管束の内側にある導管(木部)だけを通って上っていました。 The red particles rose only through the vessels (xylem) on the inner side of the vascular bundles. None rose through the phloem (outside), the epidermis or the cortex; the cortex was only where water moved sideways from the root hairs to the vessels.
How did Transpiration change when you changed Wind strength and Air humidity?→ 最初は約184粒/分でした。 It started at about 184 particles/min. With Wind strength 100 it was about 272 and with 0 about 95; with Air humidity 0% it was about 340 and with 100% about 27. Stronger wind and drier air increase transpiration.
What happened to Transpiration and Water rising speed when Open stomata was set to 0%?→ 蒸散量は 0 粒/分になり、気孔から青い粒が出なくなりました。 Transpiration dropped to 0 particles/min and no blue particles came out of the stomata. Water rising speed also fell to about 2%, almost stopping. At 100% it rose to about 262 particles/min.
On which side of the leaves were most stomata drawn?→ 気孔は主に葉の裏側(下側の表皮)にあり、表側には少ししかありません。 Most stomata are on the underside of the leaf (lower epidermis), with only a few on the upper side. Each stoma is a gap between two guard cells.

Think further

With Wind strength 100, Air humidity 0% and Open stomata 100%, Water rising speed reached 100%. Why do transpiration and the rising speed change together?→ 葉から水蒸気として出ていった分の水が、道管の中で次々に引き上げられて補われるからです。 Because the water lost from the leaves as vapor is continually replaced by water pulled up through the vessels. Transpiration is the driving force for drawing water up, so the more active it is (about 720 particles/min here), the faster the water rises, and when transpiration stops, the rise almost stops too.
In the real experiment, why do the red-stained spots on a stem cross-section appear arranged in a ring?→ セリなどの双子葉類では維管束が茎の中で輪のように並んでおり、赤い水はそれぞれの維管束の内側にある道管(木部)だけを通るからです。 In dicots such as Japanese parsley, the vascular bundles are arranged in a ring in the stem, and the red water travels only through the vessels (xylem) on the inner side of each bundle. The phloem on the outer side is not stained.

Transpiration (particles/min) and Water rising speed (%) are arbitrary units, not real amounts or speeds. The model treats dye and water as the same particles; in real experiments the dye may lag behind the water or seep into nearby cells. Use it to practice changing one variable at a time (wind, humidity, open stomata), and pair it with a real red-ink observation.
