Micelle Self-Assembly Lab

Students understand that soap molecules gather by themselves once their concentration passes the critical micelle concentration (CMC): in water they form micelles with hydrophilic heads out and hydrophobic tails in, in oil they form reverse micelles with the opposite orientation, and in an oil-water mix they line the surface of oil drops to make an emulsion.

Predict (before you touch anything)

As you slowly add more soap molecules to water, do you think they always gather together, or do they start gathering only after some number is passed?💡 情報パネルに表示される「CMCの目安」の数に注目しましょう。Pay attention to the "CMC" number shown in the information panel.
If the surroundings were all oil instead of water, which part do you think would face outward in a group of soap molecules: the head (hydrophilic) or the tail (hydrophobic)?💡 それぞれの部分は、水と油のどちらになじみやすいでしょうか。Which does each part get along with: water or oil?

Try it

In the starting state (Soap molecules 120, Amount of oil 0% (in water)), watch for a few seconds. Check which part (red hydrophilic head or yellow hydrophobic tail) lines the outside and inside of each micelle, and whether water dots get inside a micelle.
Change "Soap molecules (concentration)" to 60 → 85 → 150 → 200, wait a few seconds each time, and record the status and the "Single", "grouped" and "micelles" numbers in the information panel.
Lower the number from 200 to 60 and watch what happens to the micelles.
Set "Amount of oil" to 30% (water + oil drops = emulsion) and check the orientation of the molecules on the edge of the oil drops and the "coverage". Then set the molecules to 10, press "Stir", and watch how the coverage and the number of oil drops change.
Set the molecules to 150 and "Amount of oil" to 90% (in oil) and observe the orientation in the reverse micelles. Then lower the molecules to 50 and watch how the status changes.

Check

In water (oil 0%), what happened with 60, 85, 150 and 200 molecules?→ 60個では「分子は単独で溶けている」でミセルは0個、85個では「CMC付近(核になる分子がまだ足りない)」でまだ集合しなかった。 At 60, the status was "Molecules dissolve one by one" with 0 micelles; at 85 it was "Near the CMC (not enough molecules to start a micelle)" and no groups formed yet. At 150 about 6 micelles formed and at 200 about 9; in each case about 80-86 molecules stayed single, and the extra molecules beyond the CMC (about 80) grouped together.
In the micelles in water, which part was on the outside and which on the inside? Where were the water dots?→ 外側はすべて親水基(赤い頭部)、内側に疎水基(黄色の尾部)が隠れていた。 The outside was covered entirely by hydrophilic heads (red), and the hydrophobic tails (yellow) were hidden inside. The water dots were pushed outside the head layer and never entered the micelle core.
At 30% oil, how did the oil drops differ between many molecules and only 10?→ 分子が多いと頭部を水側・尾部を油側に向けて油滴の縁をおおい、被覆率100%で「油滴は合体しない」と表示された。 With many molecules, they covered the edge of the oil drops with heads toward the water and tails toward the oil, coverage was 100%, and the panel said "drops stay apart". With 10, coverage was below 20%, the panel said "oil drops merge", and drops split by stirring joined together again.
At 90% oil (in oil), how did the molecules gather? What happened when you reduced them to 50?→ 親水基が内側で中の水を囲み、疎水基が外側を向く「逆ミセル」ができた。 Reverse micelles" formed, with the hydrophilic heads inside surrounding water and the hydrophobic tails facing out. Reducing to 50 put the number below the CMC, the status became "Molecules dissolve one by one in oil", and there were 0 reverse micelles.

Think further

Why do soap molecules in water gather with their hydrophilic heads outside and their hydrophobic tails inside?→ 疎水基は水となじまないので、尾部どうしで集まって水にふれる部分を減らした方が安定する。 Hydrophobic tails do not mix with water, so it is more stable for them to cluster together and reduce their contact with water. With water-loving heads covering the outside, the whole cluster can stay in the water. In oil, the preferred partners are reversed, so the orientation is reversed too.
Why does washing work poorly when there is too little soap? Explain using what you saw in this simulation.→ 分子が少ないとCMCに届かずミセルができないうえ、油滴の表面をおおいきれず(被覆率が低い)、分けた油滴がまた合体してしまう。 With too few molecules, the concentration does not reach the CMC so no micelles form, and the molecules cannot cover the oil drop surfaces (low coverage), so split oil drops merge again. With enough soap, the drops are covered, stay apart, and remain spread through the water.

The model shows the CMC as a number of molecules (about 80), but a real CMC is a concentration (mol/L) that depends on the type of soap and the temperature. Micelles are drawn as flat circles, while real ones are 3D spheres, and molecule sizes and numbers are simplified. Counts fluctuate randomly, so have students wait a few seconds before reading them.
