Moths in the Sooty Forest: Industrial Melanism and Natural Selection
Students understand that because birds eat the moths that stand out most, the frequency q of the dark (melanic) allele rises along an S-shaped curve in a soot-darkened forest and the direction of selection reverses when the bark becomes light again, and that the smaller the effective population size Ne, the more genetic drift pulls q away from the expected curve.

Predict (before you touch anything)
In a forest where soot has blackened the tree trunks, how do you think the proportions of light and dark moths will change over many generations? Will the change go at the same speed the whole time?💡 鳥は幹の上で目立つ蛾を見つけやすいこと、そして黒色型が顕性(優性)であることに注目しましょう。Think about which moths a bird can spot easily on the trunk, and remember that the dark form is dominant.
If there is no selection at all (both forms are eaten with the same chance), do you think q can still change? Would a small population behave differently from a large one?💡 次の世代は、生き残った親の配偶子から「くじ引き」のように選ばれると考えてみましょう。Imagine the next generation being drawn from the survivors' gametes like a lottery.
Try it
In the starting state ("Soot cover" 80%, "Start dark freq." q0 0.03, "Effective pop." Ne 854), watch one generation. Read "Eaten: L 39% D 8%" at the top left and confirm that the bird eats mostly light moths. You may press "Pause" and then "Next gen" to advance one generation at a time.
Press the "Speed" button until it shows ×32 and run about 50 generations. In the graph on the right, watch the blue "Actual" line rise in an S shape along the orange dashed "Expected" line, and record q and the Light/Dark counts at the bottom.
When q reaches about 0.9, move "Soot cover" down to 0%. Watch white lichen appear on the trunk and the readout change to "Eaten: L 7% D 40%", and then see q fall slowly at first and then quickly.
Set "Soot cover" to 42% and check that the chance of being eaten becomes 24% for both light and dark. Set "Start dark freq." to 0.50 and "Effective pop." to its minimum of 20, press "Reset", and watch how q moves at ×32.
Set "Soot cover" to 80%, "Start dark freq." to 0.03 and "Effective pop." to 5000, press "Reset", and compare how closely the actual line follows the dashed line with the Ne = 20 run.
Check
What were the chances of being eaten for light and dark moths at 80% and at 0% soot cover?→ 80% では淡色 39%・黒色 8% で、暗い幹の上で目立つ淡色型が多く食べられた。 At 80% the light form was eaten 39% and the dark form 8%, so the light moths that stand out on dark bark were eaten more. At 0% it reversed to light 7% and dark 40%, so the dark moths that stand out on light bark were eaten more. The chance is "0.6 × |moth brightness − bark brightness|", so the larger the brightness difference, the higher it is.
Starting from q0 = 0.03 at 80% soot, how did q increase?→ 初めはゆっくり、q が 0.2〜0.7 あたりで急に、0.9 を超えると再びゆっくり増える S字の曲線になった。 It made an S-shaped curve: slow at first, fast between about q = 0.2 and 0.7, and slow again above 0.9. After about 50 generations q was about 0.9 and almost all 40 moths shown were dark.
Did the share of light moths on screen roughly match (1−q)²?→ 一致していた。 Yes. The "Light moths: seen …% / expected (1−q)²=…%" line below the graph, and the "Total light share" vs "expected" at the bottom, give nearly the same values (for example 19.9% and 19.4%). Light moths are recessive homozygotes mm, so their share is (1−q)².
At 42% soot, where both forms are eaten equally, how did q behave with Ne = 20 compared with a large Ne?→ 期待値の破線は q0 のまま水平だが、Ne = 20 では実現値が上下に大きく揺れ、±1σ帯を外れて数十世代で 1.0(「黒色アレル固定」)や 0(消失)に達した。 The expected dashed line stays flat at q0, but with Ne = 20 the actual line jumped up and down, left the ±1σ band, and within tens of generations reached 1.0 ("Dark allele fixed") or 0 (lost). With Ne = 5000 the actual line almost overlaps the dashed line. Even without selection, chance (genetic drift) changes q in a small population.
Think further
After q has risen to about 0.9, why does q fall only slowly at first when the soot is set back to 0%?→ 淡色の対立遺伝子 m は潜性なので、q が大きいと m の多くはヘテロ接合体 Mm の中にあり、黒色型として見えるため選択を受けない。 The light allele m is recessive, so when q is large most m alleles sit in Mm heterozygotes, which look dark and are not selected against. While there are few mm light moths, selection works weakly; once m becomes common enough for light moths to appear, q falls quickly. For the same reason, the last approach of q toward 1 in a sooty forest is slow.
Using this simulation, explain the difference between natural selection and genetic drift.→ 自然選択は環境(幹の明るさ)によって生存率に差があるために、決まった向きに q が変わる現象で、破線の期待値として表れ、すすを変えると向きも逆転する。 Natural selection changes q in a definite direction because survival differs with the environment (bark brightness); it shows up as the dashed expected line, and its direction reverses when the soot changes. Genetic drift makes q wander in no particular direction even without survival differences, because the next generation is a random draw of a limited number of gametes; it is larger when Ne is small and can lead to fixation or loss.
Common misconceptions
The moths turned dark themselves to match the sooty environment." In the simulation each moth's color is fixed by its genotype at birth; what changes is the allele frequency in the population, because the conspicuous form is eaten more.
Without selection, allele frequencies do not change." Even when both forms are eaten equally, in a small population q wanders by genetic drift and can even become fixed or lost.

The 40 moths on the left are a display sample; the next q is set by drawing 2Ne gametes binomially from the frequency after selection, so the fate of those 40 moths does not map one-to-one onto the change in q. Predation is a simple model proportional to the brightness difference, and both forms are equally at risk at about 42% soot. Results are random and vary from run to run (after the soot is reset, the actual line can lag well behind the dashed line), so have students try several runs.
