自然選択と遺伝的浮動(Wright–Fisherモデル)Natural Selection and Genetic Drift (Wright–Fisher Model)

生物 · 生物単元: 進化と自然選択・遺伝的浮動Natural Selection and Genetic Drift (Wright–Fisher Model) 目安 25分about 25 min
Students understand that allele frequencies change in a consistent direction under natural selection but also change by chance through genetic drift, and that drift is stronger in small populations, so even an advantageous allele can be lost.

Predict (before you touch anything)

An allele A1 that is neither helpful nor harmful (selection coefficient s = 0) makes up half of the gene pool (p = 0.5). What do you think happens to its frequency over many generations? Is there a difference between a population of 10 and a population of 2000?💡 次の世代の遺伝子は、親の遺伝子の中から「くじ引き」のように選ばれると考えてみましょう。 Think of the next generation's genes as being drawn like a lottery from the parents' genes. What tends to happen to the results when only a few draws are made?
When an advantageous allele has just appeared by mutation and is still rare (p = 0.05), do you think it will always spread through the population?💡 少数しかいない遺伝子が、たまたま次の世代に受け継がれなかったらどうなるでしょうか。What happens if, by chance, the few copies of that allele are not passed on to the next generation?

Try it

With the default settings (Start freq. p₀ = 0.50, Selection s = +0.050, Pop. size Ne = 100, "Fixed seed ON"), let it play and compare the yellow line "Actual (sel.+drift)" with the green dashed line "Deterministic (sel.)". Also watch the A1A1/A1A2/A2A2 bars at the top and the fixed/lost message.
Press "Fixed seed ON" to switch it to "Fixed seed OFF", then set Selection s to 0 and Pop. size Ne to 10. If it is paused, press "Play"; each time a run ends, press "Play" again, for 5–6 runs. Record whether each faint past-run line ends at fixation (p=1) or loss (p=0).
Set Selection s to +0.050 and Pop. size Ne to 2000, run it, and use the "drift gap" value at the top right to see how close the solid line stays to the dashed line.
Set Start freq. p₀ to 0.05, Selection s to +0.100 and Pop. size Ne to 20. Run it about 5 times with "Play" and count how often the advantageous A1 is lost and how often it becomes fixed.

Check

With the default settings (s = +0.050, Ne = 100), how did the solid line relate to the dashed line?→ 破線はなめらかに 1 に近づき、実線も全体として上向きだが、ジグザグに上下しながら破線から離れたり近づいたりする。 The dashed line rises smoothly toward 1. The solid line also trends upward, but zigzags above and below the dashed line. With seed 12345, A1 became fixed (p=1) at about generation 128.
With s = 0 and Ne = 10, what happened over repeated runs? What did the dashed line do?→ 破線は p = 0.5 のまま水平で変化しないが、実線は大きくジグザグし、多くの場合 10〜50 世代ほどで固定(p=1)か消滅(p=0)に達する。 The dashed line stays flat at p = 0.5, but the solid line zigzags wildly and usually reaches fixation (p=1) or loss (p=0) within about 10–50 generations. Which one happens differs from run to run, roughly half and half.
With Ne = 2000, what happened to the gap between the solid and dashed lines?→ 実線は破線にほぼ重なり、「差(浮動)」は 200 世代目でも ±0.01 程度と小さい。 The solid line nearly overlaps the dashed line, and the "drift gap" stays small, about ±0.01 even at generation 200. It did not reach fixation within 200 generations and showed "Reached 200 gens (not fixed)" (p about 0.99).
With p₀ = 0.05, s = +0.100 and Ne = 20, did the advantageous A1 always spread?→ 広がらないことが多い。 Usually not. In one trial, A1 was lost in 4 of 5 runs, within a few to about 15 generations, and became fixed only once. The dashed line (selection only) keeps rising slowly, but in reality the allele is often lost by chance.

Think further

Why does genetic drift have a bigger effect in smaller populations?→ 次世代の遺伝子は親の遺伝子から 2Ne 個を無作為に選んで決まる。 The next generation's genes are 2Ne copies drawn at random from the parents' genes. With few draws, the proportion can easily stray far from the expected value, like tossing a coin only a few times (the typical deviation scales roughly as 1/√(2Ne)). In a large population the random deviations cancel out and only the direction set by selection remains.
Small populations of endangered animals are said to lose genetic diversity easily. Explain why, using what you saw in this simulation.→ 小さな集団では遺伝的浮動により、有利・不利に関係なく対立遺伝子が短い世代数で固定または消滅し、集団内の対立遺伝子の種類が減っていく。 In a small population, drift drives alleles to fixation or loss within relatively few generations regardless of whether they are helpful, so the number of different alleles in the population shrinks. Even advantageous alleles can be lost, which may weaken the population's ability to adapt to environmental change.

The default "Fitness: additive" uses w11 = 1+s, w12 = 1+s/2, w22 = 1, so s > 0 favors A1 (the button switches to "Fitness: as given", a different heterozygote-advantage model, so keep it on additive in class). The model is Wright–Fisher: random mating, non-overlapping generations, no mutation or migration, with genotype bars shown at Hardy–Weinberg proportions. For repeated trials, switch to "Fixed seed OFF" or every run follows the same path; also note that moving a slider clears the faint past-run lines.
