Cross-Bridge Cycle: States of the Myosin Head

生物 · 生物単元: 筋収縮(アクチンとミオシン)Cross-Bridge Cycle: States of the Myosin Head 目安 20分about 20 min
Students understand that a single myosin head cycles through ATP binding (detachment) → ATP hydrolysis (re-cocking) → weak binding → Pi release (power stroke) → ADP release (rigor), pulling actin in the same direction each cycle, and that Ca²⁺ controls whether binding can happen while ATP controls detachment and the speed of the cycle.

Predict (before you touch anything)

When the myosin head pulls on actin (the power stroke), which of ATP, ADP, or Pi do you think leaving the head is the trigger?💡 ATP ATP is split into ADP and Pi on the head. After splitting, do the two leave at the same time?
If there is no ATP at all, do you think the myosin head will stay attached to actin or stay detached?💡 サイクルの中で ATP が最初に何をする役割なのかを考えましょう。Think about what ATP's first job is in the cycle.

Try it

With the defaults ("Ca²⁺ level" 70, "ATP level" 70), watch for about 20 seconds. Follow which of steps 1–5 is highlighted, the head angle (90°/45°), the red "one stroke" arrow, and the red marked actin subunit. Write down the order of the states.
Record the "Strokes: … (actin moved ≈ … nm)" readout below the state text, and check in which direction and how far actin moves with each stroke.
Press "Reset" and set "Ca²⁺ level" to 10. Observe the tropomyosin (pink strand), the color of the binding-site dots, and the state and angle where the head stops. Then raise "Ca²⁺ level" to 30 or more and watch what changes.
Return "Ca²⁺ level" to 70, press "Reset", and set "ATP level" to 0. Observe in which state and at what angle the head stops.
After "Reset", run with "ATP level" at 10 and then at 100 for 30 seconds each, and compare the number of strokes. Also watch how fast the "Waiting for ATP" percentage in state 5 rises.

Check

With enough Ca²⁺ and ATP, in what order did the head go through the states, and where did its angle change?→ 1 ATP結合→アクチンから離れる → 2 ATP加水分解(ADP·Pi)で頭部が 45° から 90° に直立(再加力)→ 3 露出した結合サイトに弱く結合 → 4 Pi遊離で 90° から 45° へ倒れる(パワーストローク)→ 5 ADP遊離→強直状態、そして再び 1 に戻った。1 ATP binds → detaches from actin → 2 ATP is hydrolyzed (ADP·Pi) and the head swings from 45° up to 90° (cocked) → 3 weak binding to an exposed site → 4 Pi is released and the head tilts from 90° to 45° (power stroke) → 5 ADP is released → rigor state, then back to 1.
How did actin move with each power stroke?→ 毎回同じ向き(右向き →)に動き、逆向きには戻らなかった。 It moved in the same direction (to the right, →) every time and never moved back. Each stroke added about 10 nm of relative displacement; with Ca²⁺ and ATP at 70 there were about 4 strokes (≈40 nm) in 20 seconds.
With Ca²⁺ at 10, in which state did the head stop, and why?→ 「2 再加力状態で待機(Ca²⁺不足)」で、頭部は 90° に起き上がり ADP·Pi を持ったまま止まった。 It stopped at "2 Waiting in cocked state (low Ca²⁺)": upright at 90° and still holding ADP·Pi. Troponin has no Ca²⁺, so tropomyosin covers the binding sites (gray dots) and the head cannot bind even with ATP present. Raising Ca²⁺ to 30 or more moved tropomyosin into the groove, the dots turned green, and the head went on to state 3.
What happened with ATP at 0, and how did ATP at 10 compare with 100?→ ATP=0 では「5 強直状態で停止(死後強直)」となり、頭部は 45° のままアクチンに結合して離れず、ストローク回数も増えなかった。 With ATP = 0 it showed "5 Stuck in rigor (rigor mortis)": the head stayed bound to actin at 45° and the stroke count stopped increasing. With ATP = 10 the wait in state 5 got long and there were about 3 strokes in 30 s; with ATP = 100 there were about 6. Less ATP means a slower cycle.

Think further

Explain, using ATP's two roles, why muscles lock up stiff when ATP runs out.→ ATP ATP (1) binds to the head and makes it let go of actin, and (2) provides energy, through hydrolysis, to re-cock the head to 90°. Without ATP, step (1) cannot happen, so the head stays bound to actin in the rigor state after the power stroke. When many heads are locked like this, the muscle becomes stiff (rigor mortis).
One stroke of one head moves actin only about 10 nm. How can a whole muscle still shorten a lot?→ ストロークは常に同じ向きで、頭部がサイクルをくり返すたびに変位が積み重なる(シミュレーションでも回数×約10 nm)。 The strokes are always in the same direction, so the displacement adds up each time the head repeats the cycle (in the simulation, strokes × about 10 nm). On top of that, each thick filament has many heads and a muscle fiber has many sarcomeres in series, so the small steps add up to a large contraction.

In this model the binding sites are simply all open or all covered depending on whether Ca²⁺ is at least 30, a simplification of the real graded regulation. The ATP level is a relative 0–100 value that affects only the wait for ATP binding in state 5; the other steps take fixed times. Only one head is shown and the myosin is fixed while actin moves, so pairing this with the sliding-filament sarcomere simulation helps students connect the molecular cycle to sarcomere shortening.
