Membrane close-up: ion channels and the action potential

Membrane close-up: ion channels and the action potential 目安 20分about 20 min
By matching channel movements in the membrane to the voltage record, students understand that an action potential arises when, past the threshold, Na+ channel activation gates open and Na+ flows in, then inactivation gates close and K+ flows out through K+ channels that opened later, and that K+ channels closing slowly causes a brief hyperpolarization.

Predict (before you touch anything)

When Na+ channels open, which way do you think Na+ moves: from outside to inside the cell, or from inside to outside? What happens to the membrane potential then?💡 画面上部の細胞外と下部の細胞内で、Na+(緑の粒)とK+(オレンジの粒)がどちらに多いかを見てみましょう。Look at whether Na+ (green particles) and K+ (orange particles) are more common outside the cell (top) or inside the cell (bottom).
After an action potential, do you think the membrane potential returns straight to −70 mV?💡 Na+チャネルとK+チャネルは、同じ速さで開いたり閉じたりするでしょうか。Do Na+ channels and K+ channels open and close at the same speed?

Try it

Set "Playback speed" to 0.25× and "Stimulus strength" to 10 mV, then press "Stimulate". Watch whether the line in the voltage record reaches Threshold -55mV and what the Na+ channel "Activation gate" label shows.
Set "Stimulus strength" to 14 mV and 16 mV, pressing "Reset to resting potential" before each stimulus, and compare whether a spike occurs. Then stimulate at 30 mV and compare the peak height.
Stimulate at 20 mV. As the phase bar moves Rest → Depolarization → Peak → Repolarization → Hyperpolarization → Recovery, compare the Na+ channel's activation gate and inactivation gate (ball), the K+ channel gate, the Na+ influx ↓ and K+ efflux ↑ arrows, and the "Na+ permeability" and "K+ permeability" shading on the graph.
Stimulate at 30 mV, and press "Stimulate" again just after the peak while the red "Absolute refractory" label is shown. Next, stimulate again after enough time has passed (about 10 ms or more after the first stimulus) and compare.
Check that the Na+/K+ pump's "Always on: ○ cycles" count keeps increasing even when there is no stimulus, then open and read "Explanation (how the pump and gates work)".

Check

What did the membrane potential do when the stimulus strength was 10 mV, 14 mV, 16 mV and 30 mV?→ 10 mV At 10 mV the potential only rose to about −60 mV, and at 14 mV to about −57 mV; "Below threshold: no spike, fades" appeared and it returned to −70 mV. At 16 mV and 30 mV the threshold (about −55 mV) was crossed and an action potential with a peak of about +35 to +39 mV occurred. A stronger stimulus hardly changes the peak (the all-or-none law).
During depolarization (rise) and repolarization (fall), which gates opened or closed, and which way did the ions move?→ 脱分極では Na+チャネルの活性化ゲートが開き、Na+ が細胞外から細胞内へ流入(下向きの矢印)して膜電位が急上昇した。 In depolarization, the Na+ channel activation gate opened and Na+ flowed from outside to inside (downward arrow), so the potential rose sharply. Near the peak the inactivation gate (ball) closed and Na+ inflow stopped; the K+ channel gate opened later and K+ flowed from inside to outside (upward arrow), so the potential fell. On the graph, the K+ permeability hump comes after the Na+ permeability hump.
After repolarization, how low did the membrane potential go? What were the K+ channels doing then?→ 約 −86 mV まで下がり(過分極)、静止膜電位 −70 mV より低くなった。 It dropped to about −86 mV (hyperpolarization), below the resting potential of −70 mV. At this point the K+ channel gate was still "open" or "half open"; because it closes slowly, K+ kept flowing out. Then the potential slowly returned to −70 mV (Recovery).
What happened when you stimulated again just after the peak, and when you stimulated again about 10 ms or more later?→ ピーク直後は「不応期のため再発火しない」と表示され、30 mV で刺激しても新しい活動電位は発生しなかった(Na+チャネルが不活性化中の絶対不応期)。 Just after the peak, "Refractory: no new spike" appeared and no new action potential occurred even at 30 mV (absolute refractory period, while Na+ channels are inactivated). When stimulated about 10 ms or more later, an action potential of about +39 mV occurred again.

Think further

If the Na+ channel had no inactivation gate and stayed open, what do you think would happen to the membrane potential?→ Na+ の流入が止まらないため、膜電位は高い値(Na+の平衡電位 +50 mV 付近)にとどまり、なかなか再分極できない。 Na+ inflow would not stop, so the potential would stay high (near the Na+ equilibrium potential of about +50 mV) and could hardly repolarize. There would also be no refractory period, so excitation could go backward or repeat. The inactivation gate keeps each action potential short and keeps conduction one-way.
The Na+/K+ pump is always working, but does it directly make the fast rise and fall of the action potential? What is the pump's role?→ 活動電位の速い変化は、電位依存性チャネルを通ってイオンが濃度差に従って受動的に流れることで起こり、ポンプが直接つくるのではない。 The fast changes come from ions flowing passively down their concentration gradients through voltage-gated channels, not directly from the pump. The pump uses ATP to move 3 Na+ out and 2 K+ in (3:2), keeping the concentration difference (more Na+ outside, more K+ inside) over the long term. This difference is what makes repeated action potentials possible.

The simulation solves Hodgkin–Huxley-type equations numerically (resting potential −70 mV, Na+ equilibrium potential +50 mV, K+ equilibrium potential −87 mV), and a stimulus is modelled as an instant jump of the membrane potential by the chosen number of mV. The pump and leak channels are combined into one leak current that holds the rest at −70 mV, so the pump animation is a schematic showing that it is always running. The threshold is about −55 mV: 14 mV does not fire, while 15–16 mV does. You may also point out that the hyperpolarization here reaches about −86 mV, a little deeper than the roughly −80 mV often shown in textbooks.
