Neuron Action Potential Simulator

Students understand that an action potential occurs, always at the same size, only when the threshold is crossed (the all-or-none law); that Na⁺ influx followed by delayed K⁺ efflux produces the rising phase, falling phase and hyperpolarization; and that local currents carry the impulse in one direction at a speed set by axon diameter and myelination.

Predict (before you touch anything)

If you make the stimulus gradually stronger, how do you think the height (peak potential) of the recorded action potential will change?💡 グラフに常に引かれている2本の水平線(−70 mV と −55 mV)が何を表しているかに注目しましょう。Think about what the two horizontal lines always drawn on the graph (−70 mV and −55 mV) stand for.
What do you think happens to the conduction speed when the axon is made thicker, and when it is wrapped in myelin?💡 髄鞘は電気を通しにくい膜で、軸索のところどころにすき間(ランビエ絞輪)があります。Myelin is a layer that does not conduct electricity well, and it has small gaps (nodes of Ranvier) along the axon.

Try it

Press "Reset". Without any stimulus, check that graph (B) Membrane potential stays steady on the Resting −70 line, and that in the P1 membrane (zoom) the Na⁺/K⁺ pump (3Na⁺ out · 2K⁺ in) keeps working.
Set "Stimulus strength" to 30% and press "⚡ Stimulate". Then stimulate at 50%, 60% and 100%, and compare the peak and minimum values in the "Experiment log (recorded at P1)".
Set "Animation speed" to 0.3× and stimulate at 70%. While "P1 state" changes from Rising phase → Falling phase → Hyperpolarization, compare how the Na⁺ channel and K⁺ channel open and close in the P1 membrane (zoom) with the "Open" curves (Na⁺ and K⁺ (opens later)) under the graph.
In the axon of panel (A), watch the "Local current→" label as the impulse moves P1 → P2 → P3, and the "Abs. refractory" marker on the region the impulse has just passed.
Set "Animation speed" back to 3.0×. Set "Axon diameter (thin–thick)" to 1, 5 and 10 µm, stimulate each time, and record "Estimated speed" and "Measured (P2→P3)". Then tick "Myelinated (saltatory conduction)" and repeat with the same diameters (at 0.2× it is easy to see the nodes fire one by one).

Check

What happened when the stimulus strength was 30%, 50%, 60% and 100%?→ 30% では最大約 −62 mV、50% では約 −57 mV まで脱分極しただけで減衰し、活動電位は発生しなかった。 At 30% the membrane only depolarized to about −62 mV, and at 50% to about −57 mV, then faded with no action potential. At both 60% and 100%, the same action potential appeared, with a peak of about +36.6 mV and a minimum of about −80.3 mV. The boundary is around 55–58%, depending on whether the threshold (−55 mV) is reached.
During the rising and falling phases, which channels were open, and which way did the ions move? When did the K⁺ channels open?→ 上昇相では Na⁺チャネルが開いて Na⁺ が細胞内へ流入し、膜電位が約 +37 mV まで上がる。 In the rising phase, Na⁺ channels open and Na⁺ flows into the cell, raising the potential to about +37 mV. In the falling phase, the Na⁺ channels are inactivated, the K⁺ channels open, and K⁺ flows out of the cell. In the "Open" graph, the K⁺ curve rises later than the Na⁺ curve and closes slowly.
After the falling phase, what did the membrane potential do before returning to −70 mV?→ いったん約 −80 mV まで下がり(過分極)、そのあとゆっくり −70 mV に戻った。 It first dropped to about −80 mV (hyperpolarization) and then slowly returned to −70 mV. Hyperpolarization happens because the K⁺ channels close slowly; afterwards the Na⁺/K⁺ pump and leak bring the membrane back to the resting potential.
How did the measured conduction speed (P2→P3) change with axon diameter and myelination?→ 無髄では 1 µm で約1.0 m/s、5 µm で約2.1 m/s、10 µm で約2.9 m/s と、太いほど速くなった。 Without myelin, the speed was about 1.0 m/s at 1 µm, about 2.1 m/s at 5 µm and about 2.9 m/s at 10 µm, so thicker axons are faster. With myelin, it was about 6 m/s at 1 µm, about 28 m/s at 5 µm and about 56 m/s at 10 µm, much faster than an unmyelinated axon of the same diameter. In the myelinated axon, only the nodes of Ranvier fired, one after another.

Think further

Local currents flow both forward and backward from an excited region, so why does the impulse not travel back the way it came?→ 興奮が通り過ぎた直後の部分は Na⁺チャネルが不活性化した絶対不応期にあり、局所電流を受けても再び興奮できないから。 The region the impulse has just passed is in the absolute refractory period, when its Na⁺ channels are inactivated, so it cannot fire again even if local current reaches it. Therefore only the unexcited membrane ahead is triggered, and the impulse moves in one direction.
If the size of an action potential never changes, how might our nervous system tell weak and strong stimuli (for example mild and strong pain) apart?→ 1本のニューロンの活動電位の大きさは同じなので、強い刺激は活動電位が発生する頻度(回数)の増加や、閾値の異なる多くのニューロンが興奮すること(興奮するニューロンの数)によって伝えられる。 Because each neuron's action potentials are all the same size, a stronger stimulus is signaled by more frequent action potentials and by more neurons (with different thresholds) firing. This simulation shows one action potential per stimulus, so it does not show frequency coding directly.

Channel gating is drawn with schematic curves based on time since firing, not the real Hodgkin–Huxley equations, and the estimated speed uses rough rules (unmyelinated ≈ √diameter, myelinated ≈ 6 × diameter in µm). The animation is slowed so that 1 ms ≈ 1 second at 1.0×, so show saltatory conduction at 0.1–0.3×. Near the threshold (about 55–58%) whether the axon fires can vary between trials, so use values a little apart, such as 50% and 60%, to explain the threshold.
