MHC Antigen Presentation: What Does a CTL Attack?

生物基礎 · 生物単元: 免疫のしくみ(自然免疫と獲得免疫)MHC Antigen Presentation: What Does a CTL Attack? 目安 20分about 20 min
Students understand that helper T cells are activated by recognizing antigen presented on MHC class II by dendritic cells, that with their cytokine help, killer T cells (CTLs) destroy, one by one, only infected cells presenting the same antigen on MHC class I, and that B cells become plasma cells whose antibodies neutralize free viruses.

Predict (before you touch anything)

Which cells in the body do you think killer T cells (CTLs) attack: the virus itself, infected cells, or uninfected cells?💡 T細胞は、細胞の表面にあるMHCという「台」にのせられた抗原の断片を見て判断します。T cells decide by looking at fragments of antigen placed on a "stand" called MHC on the cell surface.
What do you think happens to the CTL and B cell responses if helper T cells do not work?💡 ヘルパーT細胞の名前の「ヘルパー」が、だれを何で助けるのかを考えよう。Think about whom the "helper" in the name helps, and with what.

Try it

With the default settings (A. Virus infection rate 40%, B. Antigen presentation 60%, C. Cytokine amount 60%), press "Reset" then "Start". In the tissue on the left, watch infected cells (numbered, red) put antigen A ◆ on MHC I (1 stalk); in the lymph node on the right, watch dendritic cell 1 put antigen A on MHC II (2 stalks) and present it to the Helper T.
Compare the Helper T's reaction and the log messages when dendritic cell 2 (antigen B ■ on MHC II) touches it and when dendritic cell 1 touches it.
Follow the CTLs as they check cells one at a time, and use the log to tell apart cells marked "no attack" (uninfected, MHC mismatch, free virus, etc.) from those "destroyed". Also read the "Destroyed" and "Neutralized" counts in the status line.
  4. 「B. Set "B. Antigen presentation" to 20% (below the 30% threshold), press "Reset" then "Start", run at Speed x4, and see what happens to the Helper T and the CTLs.
Return B to 60%, set "C. Cytokine amount" to 0%, 20% and 100%, pressing "Reset" then "Start" each time, and compare the number of active CTLs, destroyed cells and neutralized viruses at about 60 s.

Check

When did the Helper T become activated? What happened with dendritic cell 2?→ 樹状細胞1がMHC IIに抗原Aをのせて提示したとき(提示量60%≥閾値30%)に活性化した。 It was activated when dendritic cell 1 presented antigen A on MHC II (presentation 60% ≥ threshold 30%). Dendritic cell 2 carried the unrelated antigen B on MHC II, so the log said "antigen does not match, so no response" and it was not activated.
Which cells did the CTLs destroy, and what did they not attack?→ 破壊したのは、MHC Iに抗原Aを提示している感染細胞だけで、1個ずつ接着してパーフォリン・グランザイムで破壊した。 They destroyed only infected cells presenting antigen A on MHC I, binding to them one at a time and using perforin and granzyme. They did not attack uninfected cells (only MHC I + self peptide), MHC-mismatched cells, free viruses (which have no MHC), or cells just infected that had not yet put antigen on MHC I.
What happened when antigen presentation was set to 20%?→ ログに「提示量20%<閾値30%→認識できず未活性化」と出て、ヘルパーTは活性化しなかった。 The log said "presentation 20% < threshold 30% → not recognized, stays inactive", and the Helper T was not activated. So no cytokines were released, there were 0 active CTLs, and the B cell stayed undifferentiated; after 48 s there were 0 destroyed and 0 neutralized, and free viruses had grown to 14.
How did the results differ with cytokine amounts of 0%, 20% and 100%?→ 0%ではヘルパーTが活性化しても支援が出ず、CTLもB細胞も応答しなかった。 At 0%, even though the Helper T was activated, no help was sent and neither CTLs nor the B cell responded. At 20%, there were 2/2 active CTLs at about 60 s and infected cells still remained. At 100%, the CTLs increased to 4 and by about 60 s infected cells and free viruses were both 0, all cleared (numbers vary a little each run).

Think further

Why is it important for the body that CTLs destroy only infected cells, one at a time, without harming nearby cells?→ ウイルスは細胞の中で増えるので、感染細胞を壊すことで増殖の場をなくせる。 Viruses multiply inside cells, so destroying infected cells removes the place where they multiply. Destroying healthy cells too would damage the tissue. By recognizing infected cells from the antigen fragments on MHC I, only the cells that need removing are eliminated.
In HIV infection (AIDS), helper T cells are infected and their numbers fall, so immunity weakens. Explain why, based on the results of this simulation.→ サイトカイン量0%やヘルパーTが活性化しない条件のように、ヘルパーTの支援がないとCTLは活性化せず、B細胞も形質細胞にならない。 As in the runs with cytokine 0% or with an inactive Helper T, without helper T support the CTLs are not activated and the B cell does not become a plasma cell. Both cell-mediated and humoral immunity weaken, so the body becomes vulnerable even to pathogens that are normally harmless.

In this model, CTL activation is shown only through cytokine help from the Helper T; presentation on MHC I by dendritic cells (cross-presentation) is not shown. MHC-mismatched cells are not attacked to illustrate that T cells recognize antigen only on self MHC (MHC restriction); in reality, cells carrying another person's MHC, as in transplants, can be targets of rejection. Infection and movement are random, so numbers vary a little each run.
