Adrenaline Raises Blood Sugar: the cAMP Cascade
Students understand that adrenaline does not enter liver cells but binds to receptors on the cell membrane, that the signal is amplified in the order G protein → adenylyl cyclase → cAMP → PKA → phosphorylase kinase → glycogen phosphorylase, and that glucose released from broken-down glycogen enters the blood and raises blood sugar.

Predict (before you touch anything)
Adrenaline is a water-soluble hormone. When it reaches a liver cell, do you think it works by entering the cell, or from the outside?💡 細胞膜はおもに脂質でできています。 The cell membrane is made mainly of lipids. Think about whether a water-soluble substance can pass through it.
How do you think a very small amount of adrenaline can make the liver release a large amount of glucose in a short time?💡 細胞の中で信号が何段階も伝わっていくとき、各段階で分子の数がどうなるかに注目しよう。Pay attention to what happens to the number of molecules at each step as the signal is passed along inside the cell.
Try it
Observe with the default settings (Adrenaline released 4 per pulse, Amplification per step c = 3, Glycogen stored 18 granules). Watch the orange adrenaline molecules in the blood vessel bind to the "Receptor" on the outside of the liver cell membrane and disappear, and watch the dots in the cascade boxes increase from step to step.
Watch particles leave the glycogen granules, change color as "glucose-1-phosphate → glucose-6-phosphate → glucose", pass through the "Transporter" into the blood vessel, and follow the "Blood glucose" graph at the top (release lasts about 12 s and repeats every 40 s).
Change "Amplification per step, c" to 2 and then 5, pressing "Reset" each time, and compare the "Amplification c⁴" value, the "Active phosphorylase (relative)" value, and how much blood sugar rises.
Return c to 3, set "Adrenaline released" to 1 and then 10, press "Reset", and compare the number of "Bound receptors" and the steady-state value of active phosphorylase.
Set "Glycogen stored" to the minimum (3 granules), press "Reset", and watch the blood glucose graph after "Glycogen left" reaches 0.
Check
How far into the liver cell did the adrenaline molecules go?→ 細胞膜の外側にある受容体に結合したところで消え、細胞の中には入らなかった。 They disappeared after binding to receptors on the outside of the cell membrane and never entered the cell. Only the signal is passed inside, from the receptor to the G protein.
When c was set to 2, 3 and 5, what were the "Amplification c⁴" values, and how did blood sugar rise?→ c⁴ はそれぞれ 16、81、625。 c⁴ was 16, 81 and 625. Because active phosphorylase per receptor is multiplied by c⁴, one release raised blood sugar to about 96 mg/dL at c = 2, about 110 mg/dL at c = 3, and close to 140 mg/dL at c = 5 (values vary a little each run).
In what order did the particles made from glycogen change before entering the blood?→ グリコーゲン → グルコース-1-リン酸 → グルコース-6-リン酸 → グルコースの順で、最後に輸送体を通って血管へ出た。 Glycogen → glucose-1-phosphate → glucose-6-phosphate → glucose, and finally out through the transporter into the blood vessel. It never ran backwards.
What happened with only 3 granules of glycogen stored? And what happened to blood sugar after release stopped?→ 分泌中なのに10秒ほどでグリコーゲン残量が 0/30 になり、それ以上グルコースが出なくなって血糖の上昇は 105 mg/dL 前後で止まった。 Glycogen left reached 0/30 after about 10 s even though adrenaline was still being released, so no more glucose came out and blood sugar stopped rising at around 105 mg/dL. In every setting, once release stopped, blood sugar slowly returned toward the normal range (baseline 90 mg/dL).
Think further
What is the advantage of passing the signal through many steps from the receptor to glycogen phosphorylase?→ 各段階で分子の数が増えるので、増幅率が掛け算で大きくなる(c=3 の4段階で 81 倍)。 The number of molecules increases at each step, so the amplification multiplies (81 times over four steps with c = 3). This lets a tiny amount of adrenaline break down a large amount of glycogen quickly, raising blood sugar fast to supply energy, for example in an emergency.
In this work, reducing the glycogen store limited the rise in blood sugar. What would you expect if adrenaline were released after long exercise or a long time without eating?→ 肝臓のグリコーゲンがすでに減っているため、信号がいくら増幅されても分解できる材料が少なく、血糖はあまり上がらない。 Liver glycogen is already low, so no matter how much the signal is amplified there is little to break down, and blood sugar rises only a little. The body must rely on other sources, such as breaking down fat.
Common misconceptions
All hormones work by entering the cell." Water-soluble hormones such as adrenaline cannot cross the cell membrane; they bind to membrane receptors and act through signaling inside the cell (such as cAMP).
One adrenaline molecule makes one glucose molecule." Because the cascade amplifies the signal over several steps, a single binding event produces many glucose molecules.

This is a simplified model: only the four steps from cAMP onward multiply by c, while the G protein and adenylyl cyclase are ×1 (in reality one receptor activates many G proteins, and several cAMP molecules bind to activate one PKA). There are only 4 receptors and binding is random, so values vary a little each run. The return of blood sugar after release stops (the work of insulin and others) is simplified as an exponential decay toward a baseline of 90 mg/dL.
