How Antibody Titer Changes: Memory Cells and the Secondary Response
Students understand that only B cells whose receptors fit the antigen are selected and multiply (clonal selection), and that because memory cells remain, the secondary response to the same antigen has a shorter lag, a higher antibody titer, and lasts longer than the primary response.

Predict (before you touch anything)
When the same antigen enters the body a second time, how do you think the antibody titer graph will compare with the first time? Predict three things: the time until it starts rising, the highest value, and how long it lasts.💡 1回目の応答が終わったあとも、体内に何が残っているかを考えよう。Think about what remains in the body after the first response is over.
If the surface shape (epitope) of the antigen has changed by the second time, do you think the antibodies made the first time can still bind to it?💡 抗体と抗原の結合は「鍵と鍵穴」にたとえられます。Antibody–antigen binding is often compared to a lock and key.
Try it
Play with the default settings (Antigen dose 60%, Re-exposure: day 25, Mutation rate 0%). In the body view on the left, watch how only one of the six B cell types, the one that fits the antigen's shape, glows and multiplies, and how Y-shaped antibodies released by plasma cells bind to antigens until they are "Removed".
On the graph on the right, compare how the antibody titer rises after the first exposure and after the re-exposure (day 25), and watch the number of "Memory cells" in the body view.
Press "Compare 1st & 2nd" and read the two curves along with the lag, peak and duration values at the top of the graph.
Still in the comparison view, set "Antigen dose" to 10% and then 100%, and see how the 1st and 2nd values and the number of memory cells ("… formed in primary" in the status line) change.
Return "Antigen dose" to 60%, raise "Mutation rate" to 50% and 100% and read the comparison values, then press "Close comparison", play it, and watch blue antibodies fail to attach to the orange mutant antigens (a red × appears).
Check
With the default settings, what were the lag, peak and duration of the primary and secondary responses?→ 一次はラグ約5.1日・ピーク34・持続8日、二次はラグ約1.9日・ピーク152・持続57日。 Primary: lag about 5.1 days, peak 34, lasting 8 days. Secondary: lag about 1.9 days, peak 152, lasting 57 days. The maximum production rate was 11 for the primary vs 80 (per day) for the secondary, so the secondary response is faster, higher and longer.
After the first exposure, which of the six B cell types multiplied?→ 受容体のくぼみが抗原の三角形の突起(エピトープ)とぴったり合う1種類だけが黄色く光って増殖し、ほかの形のB細胞は増えなかった(クローン選択)。Only the one type whose receptor notch exactly fits the antigen's triangular bump (epitope) glowed yellow and multiplied; B cells with other shapes did not (clonal selection).
When Antigen dose was set to 10% and to 100%, how did the number of memory cells formed in the primary response and the secondary response change?→ 10%では記憶細胞6個で、一次はラグ7.0日・ピーク14、二次はラグ3.0日・ピーク88。 At 10%, 6 memory cells formed; the primary had a lag of 7.0 days and peak 14, and the secondary a lag of 3.0 days and peak 88. At 100%, 20 memory cells formed; the primary had a lag of 3.5 days and peak 50, and the secondary a lag of 1.0 day and peak 200. The more memory cells, the faster and higher the secondary response.
What happened to the secondary response when Mutation rate was set to 50% and 100%?→ 50%ではピークが77に下がり持続43日に短くなった。 At 50%, the peak fell to 77 and the duration shortened to 43 days. At 100%, it became lag 5.1 days, peak 34, lasting 8 days, the same as the primary response. Existing antibodies cannot bind the mutant antigen (red ×), so a different B cell is newly selected and the response starts from scratch.
Think further
Why are vaccines often given in two or more doses? Explain using the results of this simulation.→ 1回目の接種で抗原に合うB細胞が選ばれ、記憶細胞ができる。 The first dose selects B cells that fit the antigen and creates memory cells. At the second dose, the memory cells quickly become plasma cells, giving a much higher and longer-lasting antibody titer than the primary response. This prepares the body to remove the real pathogen quickly when it enters.
Why do people need a new flu vaccine every year?→ インフルエンザウイルスは表面の形(エピトープ)が変わりやすい。 The influenza virus changes its surface shape (epitope) easily. As seen when the mutation rate was raised, a changed shape no longer fits old antibodies and memory cells, so the secondary response weakens (at 100% mutation it is the same as a primary response). So a vaccine matched to the currently circulating strain is needed.
Common misconceptions
When an antigen enters, all B cells multiply at once." In fact, only B cells whose receptor shape fits the antigen are selected and multiply (clonal selection).
Immune memory means antibodies stay in the body forever." Antibodies from the primary response drop below the detection level within weeks. What lasts is memory cells, which make lots of antibody quickly on re-exposure.

The titer is a relative value, and "lasts" is the number of days above the detection level (10). Time is strongly compressed, and antibodies drift toward nearby antigens, but whether they bind depends only on shape fit. An antigen covered by two or more antibodies is neutralized and removed shortly after. Helper T cells are not shown, so add them in discussion if needed.
