Blood Glucose Negative Feedback: Insulin and Glucagon

生物基礎 · 生物単元: 血糖値の調節とホルモンBlood Glucose Negative Feedback: Insulin and Glucagon 目安 20分about 20 min
Students understand that blood glucose is kept in the normal range (about 70–110 mg/dL) by negative feedback: when it is high, β cells in the pancreatic islets release insulin, which lowers it, and when it is low, α cells release glucagon, which raises it.

Predict (before you touch anything)

When blood glucose rises after eating sugar, which hormone do you think the pancreas releases: insulin or glucagon? What do you think happens to blood glucose after that?💡 体は血糖値を一定の範囲に保とうとします。 The body tries to keep blood glucose within a set range. If it goes too high, which way will the body try to push it back?
If the ability to release insulin becomes weak, how do you think blood glucose will change after a meal?💡 血糖値を下げるはたらきをもつホルモンが何種類あるか考えてみましょう。Think about how many hormones are able to lower blood glucose.

Try it

When the page opens, it starts playing with "Sugar per meal" at 120 mg/dL and "β-cell insulin output" at 100%, and a meal is eaten automatically at 5 min. Watch the blood glucose curve, the "Secreting:" display, the blue particles (insulin), and the liver's glycogen dots and % until about 80 min.
Press "Reset", set "β-cell insulin output" to 0%, then press "Eat sugar". Watch blood glucose until about 50 min.
Press "Reset", set the output to 30%, and press "Eat sugar". Compare how long it takes to return to the normal range with the 100% case.
Press "Reset", set the output back to 100%, and press "Exercise (uses sugar)" once. When blood glucose drops below 70 mg/dL, watch the "Secreting:" display, where the red particles (glucagon) go, and how the glycogen % changes.

Check

With 100% output and a 120 mg/dL meal, about how high did blood glucose go, and what happened after that?→ 摂取後まもなく約195 mg/dLまで上がり、インスリンが分泌されて少しずつ下がり、約40分ごろ110 mg/dLを下回った。 Shortly after the meal it rose to about 195 mg/dL, then fell gradually as insulin was released, dropping below 110 mg/dL at around 40 min. Insulin release then stopped ("Secreting: none") and glucose settled at about 90 mg/dL.
While insulin was being released, what was happening in the liver (glycogen) and in the body cells?→ 肝臓ではグルコースがグリコーゲンとして蓄えられ、グリコーゲンの粒と%が増えた(約50%→約62%)。 In the liver, glucose was stored as glycogen, so the glycogen dots and % increased (about 50% → about 62%). The body cells took in glucose (yellow particles) from the blood.
What happened to blood glucose when you ate sugar with the output at 0%? At 0% no insulin was released, and blood glucose stayed high at about 205 mg/dL, hardly falling at all. At 30% it did fall, but very slowly, and was still above 110 mg/dL even after about 90 min.
When exercise lowered blood glucose, which hormone was released, from which cells, and in which organ did it act?→ 血糖値が70 mg/dLを下回るとα細胞からグルカゴン(赤)が分泌されて肝臓に向かい、グリコーゲンが分解されて(約50%→約44%)グルコースが血液中に放出され、血糖値は約80 mg/dLまで回復した。When blood glucose fell below 70 mg/dL, α cells released glucagon (red), which travelled to the liver. Glycogen was broken down (about 50% → about 44%) and glucose was released into the blood, bringing it back to about 80 mg/dL.

Think further

Why is this system called "negative feedback"? Explain using the changes you saw in the simulation.→ 血糖値が上がるとインスリンが分泌されて血糖値を下げ、下がるとグルカゴンが分泌されて上げるというように、変化とは逆向きにはたらく結果が元の分泌を弱めるから。 Because the result works against the change and then reduces the signal that caused it: high glucose triggers insulin, which lowers glucose, and low glucose triggers glucagon, which raises it. Once glucose returned to the normal range, both hormones almost stopped and the level stayed steady.
Several hormones besides glucagon can raise blood glucose, but insulin is the only one that lowers it. Because insulin is the only way to lower blood glucose, if its release or action is too weak, glycogen synthesis in the liver and glucose uptake by cells do not happen enough, and nothing else can take over, so high blood glucose continues. This is what the 0% output run showed, when glucose stayed high.

The model is simplified: eaten sugar is absorbed within a few minutes, and glucagon is the only hormone that raises glucose (adrenaline, glucocorticoids, nervous control and loss through the kidneys are not included). An output of 0% resembles type 1 diabetes, but the model does not cover type 2 diabetes, where insulin works poorly. 120 minutes play in about 45 seconds, and one meal is eaten automatically at 5 min, so for comparisons press "Reset" and then "Eat sugar".
