(2n=4)Mitosis in a Plant Cell (2n=4)
Students understand that in plant-cell mitosis the sister chromatids of each replicated chromosome are shared equally between the two poles by spindle fibers, and that a cell plate growing from the center outward divides the cell, producing two daughter cells with the same chromosome number (2n=4) and DNA amount (2C) as the parent cell.

Predict (before you touch anything)
During interphase, before division starts, what do you think happens to the DNA amount (2C) and to the number of chromosomes (4)?💡 分裂後の2つの細胞が、どちらも元の細胞と同じ遺伝情報をもつためには何が必要か考えてみましょう。Think about what must happen so that both new cells end up with the same genetic information as the original cell.
Plant cells are surrounded by a stiff cell wall. Do you think they can split by pinching in at the surface like animal cells? If not, how might they divide the cell?💡 最後に2つの細胞の間に何がなければならないか(細胞壁)に注目しましょう。Focus on what must exist between the two new cells at the end (a wall).
Try it
Play at "Playback speed" ×1.00 and watch the bar graph on the right as the cell goes from Interphase (G1) → Interphase (S: DNA copying) → Interphase (G2). Note how the DNA bar and the chromosome bar change. Use "Pause" to freeze if needed.
Tick "Approval mode". When the display shows "Prometaphase → metaphase" and red rings (unattached kinetochores) are still visible, press "Go to anaphase" and read the warning that appears.
When "All chromosomes bioriented" appears, press "Go to anaphase" again. In anaphase, watch the chromatids move to the poles, the pole distance value (about 1.24 → 1.60), and the Left pole / Right pole bars.
When Telophase & cytokinesis begins, lower "Cell plate speed" to ×0.5 and watch the cell plate grow from the center to the edge with its % value. Confirm that division does not finish until it reaches 100% and a new wall forms.
Press "See wrong examples", view the three incorrect models one by one, and discuss what is wrong before the answer appears. Press "Exit wrong examples" to return.
Check
During interphase (S phase), how did the DNA amount and the chromosome number change?→ DNA量は2Cから4Cに倍増したが、染色体数は4本(2n=4)のままだった。 The DNA amount doubled from 2C to 4C, but the chromosome number stayed at 4 (2n=4). Each chromosome now has two sister chromatids, but the number of chromosomes does not increase.
In Approval mode, what happened when you pressed "Go to anaphase" while red rings were still visible?→ 「未接着の動原体が○個あります。 A warning appeared saying there were unattached kinetochores and that the spindle checkpoint blocks anaphase, and the cell did not enter anaphase. It could only proceed once every chromosome was attached from both poles (bioriented).
Right after anaphase began, what were the chromosome numbers and DNA amounts for the Whole cell, Left pole, and Right pole bars?→ 姉妹染色分体が分かれて独立した染色体になるので細胞全体では一時的に8本(4C)、左極と右極にはそれぞれ4本・2Cずつ分配された。Because the sister chromatids separate into independent chromosomes, the whole cell briefly has 8 chromosomes (4C), and the left and right poles each receive 4 chromosomes and 2C.
Where did the cell plate start and where did it grow to? What changed when you set "Cell plate speed" to ×0.5?→ 細胞板は細胞の中央から上下の細胞壁(外周)へ向かって伸びた。 The cell plate grew from the center of the cell toward the cell wall at the top and bottom (the edge). At ×0.5 the % rose more slowly, and cytokinesis did not complete (daughter cells 2n=4, 2C) until the plate reached 100% and the new wall formed.
Think further
If both sister chromatids of one chromosome were attached to fibers from the same pole and the cell went into anaphase, what would happen to the daughter cells? Both chromatids would go into the same daughter cell, so one cell would get 5 chromosomes and the other 3, and the genetic information would not be shared equally. The checkpoint prevents this by allowing anaphase only after every chromosome is bioriented (see wrong example 1).
Why is it important for building an organism's body that the chromosome number and DNA amount are the same in the parent cell and the daughter cells?→ 体をつくるすべての細胞が同じ遺伝情報(同じ染色体のセット)をもつことができるから。 Because it lets every cell in the body carry the same genetic information (the same set of chromosomes). DNA is copied in interphase (2C → 4C) and then split in half (2C each) during division, so 2n=4 and 2C are kept no matter how many times the cell divides.
Common misconceptions
Misconception: "When DNA is copied, the chromosome number doubles to 8." In fact, after S phase the DNA is 4C but there are still 4 chromosomes; 8 appear only in anaphase when the sister chromatids separate.
Misconception: "Plant cells divide by pinching in, just like animal cells." Because plant cells have a cell wall, a cell plate grows from the center outward to separate them.

The model is simplified to 2n=4 (a real onion has 2n=16), and in anaphase all chromatids move perfectly together at the same speed. Note that the bar graph keeps the whole cell at 8 chromosomes and 4C until the cell plate is complete (cytokinesis complete). Advanced structures such as the preprophase band and phragmoplast are shown, but for an introductory course it is enough that plant cells have no centrosomes and are divided by a cell plate.
