Meselson–Stahl Experiment Simulator
Students understand that in the Meselson–Stahl experiment, only hybrid-density DNA appears after one generation in ¹⁴N medium, and afterward the hybrid molecules stay at 2 while light DNA increases, a result explained only by semiconservative replication.

Predict (before you touch anything)
If E. coli containing only ¹⁵N is moved to ¹⁴N medium and divides once, where in the centrifuge tube do you think the DNA band(s) will appear under semiconservative replication? What about conservative replication?💡 それぞれのモデルで、新しくできた二本鎖DNAの2本の鎖がどの窒素を含むかを図にかいて考えましょう。For each model, draw which nitrogen each of the two strands in a new double-stranded DNA contains.
Try it
With "Generations grown" at G0 and "Replication model (for prediction)" set to "Semiconservative", check the position of the heavy (¹⁵N/¹⁵N) band in the left "Observed" tube against the density scale (1.724 g/cm³).
Move the "Generations grown" slider G1 → G2 → G3 → G4, and record the band positions and percentages for the observed data and the prediction, plus the breakdown of the 2ⁿ molecules shown below.
Switch to "Conservative" and compare the observation with the prediction for G1 to G3.
Switch to "Dispersive" and record the predicted band position and the "Density of molecules in this generation" value for G1 to G4.
In "Question: Which replication model matches the observations?", choose one answer and press "Check answer". If it is wrong, compare again at the generation shown in the feedback.
Check
In the observed data, which bands appeared at G1, G2 and G3, and at what percentages?→ G1 は中間(¹⁵N/¹⁴N)100%、G2 は中間50%・軽い(¹⁴N/¹⁴N)50%、G3 は中間25%・軽い75%。 G1: hybrid (¹⁵N/¹⁴N) 100%; G2: hybrid 50% and light (¹⁴N/¹⁴N) 50%; G3: hybrid 25% and light 75%. After G1 the heavy band never appeared.
In the semiconservative prediction, how did the breakdown of molecules change?→ 分子数は 2ⁿ 本(G1 2本、G2 4本、G3 8本、G4 16本)に増えるが、中間の分子は常に2本のままで、残りが軽い分子になった。 The number of molecules grows as 2ⁿ (2 at G1, 4 at G2, 8 at G3, 16 at G4), but there are always just 2 hybrid molecules and the rest are light. So the hybrid share halves each time: 100% → 50% → 25% → 12.5%.
How did the conservative prediction differ from the observation at G1?→ 保存的複製では重い50%と軽い50%の2本のバンドになり、中間バンドが現れない。 Conservative replication gives two bands, heavy 50% and light 50%, with no hybrid band. The observation is 100% hybrid, so they do not match.
How did the dispersive prediction change over the generations, and at which generation did it stop matching the observation?→ いつも1本のバンドだけで、密度が 1.717 → 1.7135 → 1.7117 → 1.7109 g/cm³ と世代ごとに軽い側へ移った。 There was always a single band, shifting toward the light side each generation: 1.717 → 1.7135 → 1.7117 → 1.7109 g/cm³. At G1 it matches the observation (100% hybrid), but at G2 it is a single band of 25% ¹⁵N molecules, which does not match the observed two bands (hybrid and light).
Think further
In semiconservative replication, why do there stay exactly 2 hybrid molecules no matter how many generations pass?→ 最初の¹⁵N二本鎖DNAの2本の鎖は、複製のたびにそれぞれ別の娘DNAに1本ずつ受け継がれ、こわれずに残る。 The two strands of the original ¹⁵N DNA are each passed intact into a different daughter DNA every time it replicates. Since there are only two ¹⁵N strands in total, there are always only two hybrid molecules containing them, and all other new molecules have two ¹⁴N strands, so they are light.
Why can't the G1 result alone distinguish semiconservative from dispersive replication? What should you look at to tell them apart?→ どちらのモデルでも G1 の分子はすべて¹⁵Nと¹⁴Nを半分ずつ含み、同じ中間の密度になるから。 In both models every G1 molecule is half ¹⁵N and half ¹⁴N, so they all have the same hybrid density. From G2 on, semiconservative replication gives two bands (hybrid and light), while dispersive replication gives one band that slowly moves toward the light side, so they can be told apart.
Common misconceptions
A band's position depends on how much DNA there is." The position (height) depends on the density of the molecules, that is, the ratio of ¹⁵N to ¹⁴N; the amount of DNA shows up as the band's thickness.
The hybrid band in generation 1 proves semiconservative replication." Dispersive replication also gives a hybrid band at G1, so only the results from G2 on show that replication is semiconservative.

Observed data cover only G0–G3; G4 shows "No data" and only the model prediction. Molecule counts are calculated from one starting molecule growing to 2ⁿ molecules, an idealized version of the absorbance bands measured in the real experiment. Having students first see that semiconservative and dispersive models agree at G1, and that G2 settles the question, deepens the discussion.
