Plasmolysis and Turgor Pressure in Plant Cells
Students understand that because the semipermeable membrane lets only water through, water moves according to the osmotic pressure difference Δπ = 2(c_in − c_out)RT; in a hypotonic solution equilibrium is reached when the wall's turgor pressure P balances Δπ, and in a hypertonic solution the cell plasmolyzes until the inside and outside concentrations are equal.

Predict (before you touch anything)
What do you think happens when a plant cell is put into pure water? Will it burst like a red blood cell?💡 植物細胞には、半透膜(細胞膜)の外側にもう一枚の丈夫な壁があります。A plant cell has another strong layer outside its semipermeable membrane (cell membrane).
If the outside solution is made a strong salt solution, which do you think moves across the membrane: water, or Na⁺ and Cl⁻?💡 「半透膜」という名前の意味を考えてみましょう。Think about what the name "semipermeable membrane" means.
Try it
In the starting state (Outside conc. c_out = 0.10 mol/L, Temp. T = 298 K), record the status shown at the top right of the canvas and the values of "Volume V/V₀", "Osmotic diff. Δπ", and "Turgor P".
Press "Pure water" and watch the cell swell, the Δπ and Turgor P bars, and the "Equilibrium" line below them.
Raise the "Outside conc. c_out" slider to 0.30 and then 0.50 mol/L, then press "Hypertonic 1.0". Watch the membrane (semipermeable membrane) pull away from the cell wall and record "c_in" and "Volume V/V₀".
Meanwhile, watch "Ions crossing membrane (total)" and "Water crossing membrane /s" to see which particles cross the membrane and which do not.
Press "Pure water" and let it reach equilibrium, then raise "Temp. T" from 298 K to 373 K and record how Δπ, Turgor P, and the volume change.
Check
In pure water (c_out = 0), did the cell burst? What were Δπ and Turgor P at equilibrium?→ 破れなかった。 It did not burst. The volume stopped at about V/V₀ ≈ 1.13 and the cell became turgid (firm from turgor). At that point Δπ = P ≈ 1314 kPa and the net water flow was zero.
What happened to the cell when c_out was 0.50 mol/L or 1.0 mol/L? Water flowed out of the cell and plasmolysis occurred: the protoplast pulled away from the wall and shrank into a rounder shape. At equilibrium c_in became equal to c_out (V/V₀ = 0.600 at 0.50 mol/L and 0.300 at 1.0 mol/L), and Δπ = P = 0 kPa. The gap between the wall and the membrane filled with outside solution.
Did Na⁺ or Cl⁻ cross the membrane?→ 通過しなかった。 No. "Ions crossing membrane (total)" stayed at 0, and only the number of bounces off the membrane increased. Only water molecules crossed the membrane; the cell wall is fully permeable, so ions did pass through it.
In pure water, how did Δπ and Turgor P change when the temperature was raised from 298 K to 373 K?→ Δπ = P Δπ = P rose from about 1314 kPa to about 1604 kPa, and the volume grew slightly from V/V₀ ≈ 1.13 to about 1.16. Osmotic pressure is proportional to the absolute temperature T (van 't Hoff's law).
Think further
Why does water stop flowing in even in pure water, when the inside osmotic pressure is still higher than outside?→ 細胞が膨らむと細胞壁が押し広げられ、壁が押し返す膨圧 P が生じる。 As the cell swells it pushes the cell wall outward, and the wall pushes back with turgor pressure P. When P equals Δπ, the push of water inward and the push back balance, so net inflow becomes zero (dV/dt = Lp(Δπ − P) = 0). Animal cells have no wall, so they lack this limit and burst easily.
Why did the volume stay at V₀ when c_out = 0.30 mol/L?→ このモデルの内液は V₀ のとき NaCl 相当 0.30 mol/L なので、外液と等張になり Δπ = 0 になる。 In this model the cell sap is 0.30 mol/L NaCl-equivalent at V₀, so the outside is isotonic and Δπ = 0. Turgor is also 0, so water moves in neither direction. With a stronger outside solution, plasmolysis begins (incipient plasmolysis).
Common misconceptions
In strong salt water, salt enters the cell and makes it shrink." Only water crosses the semipermeable membrane; Na⁺ and Cl⁻ do not. The cell shrinks because water leaves it.
Plant cells also keep taking in water until they burst." The elastic cell wall pushes back as turgor pressure, so water uptake stops when Δπ = P.

This is a simplified model: the cell sap is 0.30 mol/L NaCl-equivalent and the wall acts as a spring with modulus 10 MPa (P = E(V/V₀ − 1)); time is sped up. Real cell sap is a mixture of sugars and ions, not NaCl, and real membranes are not perfectly water-selective. The temperature goes up to 373 K; explain that this is only to show the T dependence, assuming the water does not boil.
